KRAS Inhibitor
KRAS inhibitors are direct, targeted anticancer agents—almost all of them low-molecular-weight small molecules—that bind oncogenic KRAS proteins and prevent them from propagating downstream signaling through the RAF–MEK–ERK and PI3K–AKT pathways. In contemporary usage the term refers to drugs that physically engage mutant KRAS itself, most commonly hotspot alleles such as KRAS G12C and increasingly G12D or multi-mutant variants, rather than to upstream or downstream pathway blockers such as EGFR, MET, MEK or ERK inhibitors that modulate the same signaling network indirectly. In practice, current approved agents are allele-selective covalent inhibitors that recognize the cysteine introduced by the G12C mutation, trap KRAS in its inactive GDP-bound conformation, and thereby attenuate the chronic proliferative and survival signaling that characterizes KRAS-mutant non-small cell lung cancer (NSCLC), colorectal cancer (CRC) and other solid tumors.

From a historical perspective KRAS inhibition represents the end of a four-decade “undruggable” saga. The RAS oncogenes HRAS and KRAS were among the first human cancer genes identified in the early 1980s, and activating RAS mutations are now known to drive roughly 20–30% of all human cancers, with KRAS the most frequently mutated isoform and almost universal in pancreatic ductal adenocarcinoma (PDAC). Despite intense medicinal-chemistry efforts, the small, smooth GTP-binding surface of KRAS was long thought to lack a druggable pocket. The field pivoted in 2013 when Shokat and colleagues described a previously unappreciated “switch II” allosteric pocket adjacent to the GTP-binding site that could be targeted by electrophilic fragments to form covalent adducts with cysteine at position 12 in KRAS G12C, opening the door to conformationally selective covalent inhibitors. This discovery set off an intense global race that, after nearly 40 years of negative experience, produced the first clinically successful KRAS-targeted drugs. Before 2021 there were no approved KRAS inhibitors and essentially no commercial revenue attributable to direct KRAS blockade; all treatment of KRAS-mutant tumors relied on cytotoxic regimens, immune checkpoint blockade, or pathway-level agents such as EGFR or MEK inhibitors.

The global KRAS inhibitor market is moving rapidly from an early-launch niche to a scaled targeted-oncology class. Based on APO Research’s estimates, global sales are expected to reach approximately US$0.49 billion in 2024 and grow to around US$3.18 billion by 2032, implying a strong CAGR of about 23–24% over 2025–2032. Growth is volume-driven rather than price-driven: worldwide demand is projected to increase from roughly 8.6 million to more than 78 million 120-mg-equivalent tablets over the same period, while global average net ex-factory prices gradually decline from the high-US$50s per tablet at launch to the low-US$40s by the early 2030s.
Read the full market overview
The market today is highly concentrated in a small number of tumour types and regions. Non-small-cell lung cancer (NSCLC) already accounts for the majority of KRAS inhibitor use, with colorectal cancer forming the second pillar and pancreatic plus other solid tumours contributing a small but fast-growing tail. North America remains the commercial centre of gravity, supported by high testing penetration, rapid reimbursement and comparatively high net prices. Europe scales later and at lower price points but still builds a meaningful second revenue pillar. Asia Pacific is the fastest-growing region: from a negligible base in 2021–2023, volumes expand sharply from 2024 onwards as Japan, South Korea, Australia and, critically, mainland China approve and commercialise KRAS G12C inhibitors. Because domestic products in China are priced materially below US and EU benchmarks, China’s late but rapid entry both reshapes the regional volume mix and pulls down Asia-Pacific average prices.
Competitive intensity is expected to increase as additional first-generation KRAS G12C inhibitors and next-generation molecules targeting other KRAS alleles enter the market. Amgen, Bristol Myers Squibb/Mirati, Innovent Biologics/GenFleet Therapeutics and Jacobio Pharma/Shanghai Allist Pharmaceuticals currently anchor the class, with other multinational and Chinese manufacturers developing follow-on assets. In 2024, the three largest vendors together account for well over 99% of global KRAS inhibitor revenue, but this concentration will moderate as new entrants launch, combination strategies proliferate and regional players consolidate positions in China and other emerging markets.
Report Scope
This report quantifies the global KRAS Inhibitor market in terms of revenue (US$ million) and, where applicable, sales volume (k bottles), using 2025 as the base year and providing annual historical and forecast data for 2021–2032.
It standardizes definitions of Types and Applications, harmonizes vendor attribution, and presents comparable time series by company, Type, Application, and region/country, including indicative price bands (US$/k bottles) and concentration ratios (CR5/CR10).
The outputs are intended to support strategy development, budgeting, and performance benchmarking for brand owners, manufacturers, retailers, channel partners, and investors; data are structured with consistent units and fields to facilitate integration into internal FP&A and BI systems.
